In Vivo Effects of Diosgenin on the Activity of Some Drug- Metabolizing Enzyme Systems

چکیده
This study investigated the in vivo effect of Diosgenin, isolated from Asparagus officinalis, on the total quantity of Cytochrome P450, the activity of some enzymes Ethylmorphine N-demethylase (marker for CYP3A) and Aniline Hydroxylase (marker for CYP2E1), the quantity of malondialdehyde and reduced glutathione compared to Phenobarbital in rats. Diosgenin and Phenobarbital, administered alone, increased statistically significant, compared to control, the total amount of Cytochrome P450 – with 36 % and with 85 %; the activity of EMND with 58 % and with 184 %. Diosgenin didn’t influenced the AH activity compared to Phenobarbital. Both Diosgenin and Phenobarbital had no statistically significant effect on GSH level. On the quantity of MDA, Diosgenin had no effect, while Phenobarbital increased it with 27 %. Our results showed that Diosgenin exhibited inducible effect similar to the effect of Phenobarbital on the activity of some drug-metabolizing enzyme systems.

Effects of salidroside on rat CYP enzymes by a cocktail of probe drugs
Objective(s): In this study, we aimed to evaluate the effect of salidroside on the activities of the different drug-metabolizing enzymes CYP1A2, CYP2B6, CYP2C9, CYP2D6 and CYP3A4 in rats, in which a specific probe drug was used for each enzyme. Materials and Methods: After pretreatment with salidroside, five probe drugs were simultaneously administered to rats by gavage. The given dose was 2.0 ...
متن کاملThe effect of sugar cane molasses on the immune and male reproductive systems using in vitro and in vivo methods
Objective(s): Sugar cane molasses is a commonly used ingredient in several food products. Contrasting reports suggest that molasses may have potential adverse or beneficial effects on human health. However, little evidence exists that examines the effects of molasses on the different physiological systems. This study investigated the effects of sugar cane molasses on various physiological syste...
متن کاملEffect of Honey on CYP3A4 Enzyme and P-Glycoprotein Activity in Healthy Human Volunteers
The activity of cytochrome p450 isozyme 3A4 (CYP3A4) enzyme and P-glycoprotein (P-gp) is modulated by grapefruit juice and herbal drugs. CYP3A4 is the major phase I drug metabolizing enzyme and P-gp is an ATP-dependent drug efflux pump that regulates the intestinal absorption of orally administered drugs. Honey is commonly consumed as a dietary supplement. However, its influence on human CY...

Objective(s) Several studies have reported improved response of exercised hearts to myocardial infarction (MI). This study was aimed to evaluate the preventive role of treadmill exercise and diosgenin on cardiac marker enzymes, thiobarbituric acid reactive substances (TBARS), total antioxidant status (TAS), lipids, and electrocardiographic (ECG) patterns in isoproterenol (ISO)-induced myocardi...
متن کاملThe effects of fenvalerate on hepatic and cerebral xenobiotic metabolizing enzymes in selenium and/or iodine deficient rats
Objective(s): Particularly in developing countries, selenium and/or iodine deficiencies are encountered and use of pesticides in agriculture are not well-controlled. Fenvalerate is a pyrethroid insectide used in agriculture and has applications against a wide range of pests. This study was designed to evaluate the effects of fenvalerate on hepatic and cerebral xenobiotic metabolizing enzyme act...
